How to Prepare the Culture Media? What is The Use of Culture Media ? What is the Culture Media

Culture media is the artificial nutrients to the microorganisms they are grown low to large number of bacterium and fungi. Every Microbiology Laboratories are using a Culture Mediums.

Overview of Culture Medium

Culture media is an important medium used for the artificial growth of microorganisms. It can be used to determine whether the microorganisms are identified as gram positive or gram negative. Estimate the variance associated with the same fungus. There are different types of culture medium. Microbiology laboratories use a separate medium for each sample. This makes it easy to identify the specific microorganism. Microorganisms around the world are identified in two ways, gram negative and gram positive, as well as sporing and non-sporing. Tests are performed to identify a microorganism Inoculation. By which the bacteria are Gram negative or gram positive is identified and then biochemical tests are conducted to identify the microorganism.

Culture medium is mainly of 2 types

  • Enriched medium
  • Enrichment medium

Enriched medium

This is known as solid medium and the growth of colony through this media is very good in the same way that antibiotics are given to the patient. An agars powder is boiled at 121 degrees Celsius for 15 minutes. Then the mixture is poured into petri plates in a clean (sterilized) laminar air flow. The mixture becomes solid after 2 hours.

Media Preparation Process:

  1. Take A conical Flask (1000ml).
  2. Add a Agar Powder Your Require medium and Required Quantity
  3.  Mix Well and close the cap with cotton role or aluminum foil.
  4. Check the PH with PH strip or meter.
  5. Stick the Thermal Indicator.
  6. Kept medium in Autoclave
  7. Kept Steam Indicator
  8. Start the autoclave at 121 degrees Celsius 15 Ibs Pressure.
  9. After complete the estimated time. Open the autoclave pickup the meterial again check the PH.
  10. Add the medium in petri plates.
  11. After solidify the medium  kept one plate of media in incubator for quality purpose.
